  2. Basics • Location – Southeast Asian Islands • Thorny shell, Pungent Odor • Can kill on contact With your head from High altitudes

  3. Growth • Pollination – Determined by Spider Hunters, various native bird species, and even bats.

  4. Growth • Cultivation – Not an easy task due to dangers of fruit falling on the cultivator’s head upon his inspection of the trees unless he is careful and organized.

  5. DNA Extraction • Fresh Durian Fruit Dissection  Seed Removal  Seed Dissection  Remove the Embryo  Grind the Embryo.

  6. DNA Isolation • With the proper Buffers and equipment DNA isolation is based on general experience. There WILL be mistakes in the beginning.

  7. Dye • The dye was combined with our isolated DNA simply by transferring it to a premade pebble of dye in another tube.

  8. PCR The thermal cycler uses quick changes in temperature to break and merge strings of nucleotides. High temperatures break them apart, and cool temperatures match them with their corresponding strings.

  9. Thermal Cycler • 24 Cycles • Duplicate/Clone DNA • 224 = 16,777,216

  10. Gel Electrophoresis

  11. Results???? • NONE yet due to complications with equipment and shortages.

  12. Conclusion • Our primers have not arrived, and therefore we could not come to any conclusions about the Durian family tree and what makes them so diverse.
