Genetic Sampling

  • Species:
    • Leptodeafragilis (wetlands/river mouths)
    • Pyganodongrandis (wetlands/nearshore/tribs – Lake Erie)
    • Quadrulaquadrula (wetlands/river mouths – Lake Erie)
    • Lampsilissiliquoidea/radiata (Lake St. Clair drainage only)
    • Ligumianasuta (where it can be found 1-2 swab samples/refuge)
  • Numbers (site = refuge scale):
    • Microsats – clips only:
      • Q. quadrula(at least 10, but <50/site)
      • L. fragilis(at least 10, but <50/site)
      • L. siliquoidea(at least 10, but <50/site)
    • mtDNA (m/f lineage) – sacrifice animals
      • P. grandis (15 males/site)
      • L. fragilis (15 males/site, subsampled from above) – juveniles?
    • Swabs only (endangered species)
      • L. nasuta (1-2 swabs/site)

Clipping instructions (more at methods meeting):

    • pry open shell with blunt end of scalpel/spatula (no blade!)
    • use thumb or end of pencil or small rubber stopper to wedge open shell
    • clip ~0.5 cm2 piece of mantle tissue from shell edge using surgical scissors (from basic dissection kit – keep them sharp)
    • clip from ventral-anterior side (avoid siphons)
  • Swab samples
    • as above, but sterile swab 2-3 times along foot – avoid touch mantle and siphons

  • Zanatta lab:
    • 2 grad students + 2 undergrads
    • 2 teams will join field teams as dedicated clippers/swabbers
  • Krebs lab:
    • Trevor Prescott + undergrad will assist in clipping
  • 3 genetics teams to assist other with general field work and will do clipping in field
