  1. Primer Design Project Easter has just passed. In addition to the abundance of birds and flowers around you, you begin to notice increasing numbers of rabbits. Being a big fan of The Easter Bunny, you decide to study the molecular genetics of the lagomorpha (rabbits, hares, and pikas). Because of its high phylogenetic information content you choose to focus on the mitochondrial genome. Your job (should you choose to accept it) is to design a pair of primers to amplify the complete (or as near to complete as is possible) mitochondrial gene of your choice from the lagomorph species of your choice. Keep in mind all of the considerations regarding primer characteristics that we discussed in class (length, base composition, Tm, etc.). Be sure that the primers do not amplify DNA from the most likely source of contamination: YOU!!!

  2. Once design is complete, hand in: • Identity of the lagomorph species. (Both scientific and common names) • Identity and sequence of the mitochondrial gene you are trying to amplify. • Sequence 5’-> 3’ and Tm of each primer. • Predicted length of the PCR product.
