  3. How have we achieved this • Increased use of automation • Integration of database and instruments • Changing Rotation system

  4. GT Rotation • Disorders split into 4 groups • Group 1 Triplet Repeats • Group 2 Single gene disorders • Group 3 Sequencing • Group 4 Aneuploidy Screen, Telomeres

  5. Current Rotation System • 4 months on each group Working with a lead scientist • 1 month rotation into DNA extraction

  6. This talk concentrates on areas of recently increased automation • DNA Extraction • Sequencing service

  7. DNA Extraction • DNA extracted from most blood samples by Chemagen/Perkin Elmer DNA Extraction system. • Small blood samples and all other tissues by Qiagen EZ1.

  8. Automation of DNA Extraction • Barcode reader for blood tubes and elution tubes • High risk protocol recently introduced • Encourage (some) use of Oragene kits so DNA can be processed by Chemagen to improve DNA quality.

  9. DNA Extraction data • Use Nanodrop to generate quantity and quality information on each sample. • Import this data into database

  10. Sequencing service • GT responsible for generating PCR products and sequencing. • Co-ordinating PCR products submitted by other staff.

  11. Submitting PCR products for sequencing • Details of Gene, Exon, primers etc put onto database by individual staff • This information used by GT to create plate record for Biomek and ABI 3730

  12. Pre and post PCR Biomek robots • PCR set up • Post PCR • Ampure (PCR clean up) • Sequencing set up • Cleanseq (sequencing clean up) • Elution into plate for ABI 3730

  13. Recent developments • Increased use of pre PCR robot to set up DNA dilutions and PCR reactions • Working towards the use of StarLims

  14. Summary • GTs rotate through all sections of the lab in around 18 months. • This helps with continuity of service during sickness and holidays • Increasing automation and integration avoids repeating tasks
