  2. There are several options available to generate stable cell lines depending on the purpose of the experiment.

  3. Culture Conditions • Culture conditions (passage, split rhythm, number, etc.) Stable cell lines can only be created if you have the right cell type.

  4. Design experiment • Create an experiment and select the expression vector, cell type, and transfection method. Check that the expression vector and transfection methods you choose are appropriate for your cell type.

  5. Select the G418 concentration and the cell number • Matrix titration can be used to determine the appropriate G418 concentrations and cell numbers per well. Attention to the fact that stock G418's active concentration can vary from batch-to-batch.

  6. Transfection • Transfect expression plasmid manufacturing to cells. Follow the instructions provided by your transfection device manufacturer. G418 should not be added to the culture medium right away after
