We are learning to: • Carry out aseptic technique effectively in the lab • Discuss optimal growing conditions for micro-organisms Success criteria: • Aseptic technique has been successful if there is no contamination • All micro-organisms have optimum growing conditions

Wipe surface with disinfectant to sterilise the surface Flame loop until red hot to kill bacteria Lift lid a little to prevent contamination from bacteria Flame bottle neck to stop contamination Sealing with small pieces of tape stops getting microbes in or out Sterilise equipment in an autoclave to kill bacteria

  5. Aseptic Technique • Now you will set up an experiment in your groups • Follow instructions carefully to avoid contamination of plates.

  6. What conditions would affect growth of micro-organisms? • Note answers on whiteboards • Temperature • Why? • Oxygen availability • Why? • pH • Why?

  7. What are the different types of growth media? • Broth - liquid containing nutrients for optimal growth sterilised using autoclave - micro-organism added to the liquid and kept warm to encourage growth

  8. What are the different types of growth media? • Agar • solid growth media • Can be in a petri dish or bottle • Bottle usually used to make a slope • Agar can be altered to suit what is being cultured
