  1. First Annual Meeting

  2. CTSI Project Development Team:Benefits a Young Investigator Kimberly K. Buhman, Ph.D. Assistant Professor of Foods and Nutrition Purdue University

  3. CLD CLD = Cytoplasmic Lipid Droplet The Process of Dietary Fat Absorption Enterocyte Plasma TG CM Via Lymph CM CM CM Intestinal lumen CM DG +FFA MG +FFA TG CM CM = Chylomicron Long Term Goal: to identify novel factors that differentially regulate intestinal triglyceride metabolism and may serve as targets or methods for preventive and therapeutic interventions for obesity, CHD, and diabetes.

  4. D C 50 µm 20 µm Coherent Anti-Stokes Raman Scattering Microscopy (CARS) Nonlinear Vibrational Imaging Technique with Sub-micron Resolution • Advantages • Chemical specificity • Image LIPIDS in live cells and tissues without labeling • - Strong signal from molecules enriched in C-H bonds • Can combine with other imaging techniques Ji-Xin Cheng, Sunney Xie, J. Phys. Chem.(feature article), 2004, 108, 827. Ji-Xin Cheng, Applied Spectroscopy (focal point article), 2007, 61:197A.

  5. Cytoplasmic Lipid Droplets Observed in Enterocytes of Mice Fed a High Fat Diet Lipid accumulation in mice fed a chronic high fat diet HIGH FAT 50 µm 5 µm LOW FAT 50 µm 5 µm

  6. Idea  Funded Project • Drafted specific aims page for grant and identified a potential funding mechanism • Discussed with Project Development Team (PDT) mentor • Meeting set up by PDT mentor • Presented my idea and aims to six, senior faculty members • Significant, successful grant writing experience • Modified specific aims based on feedback • Focus • Sell • Resources • Wrote grant • Submitted • FUNDED

  7. American Heart Association Scientist Development Grant, National Center, 4 year award Title: Regulation of Intestinal Triglyceride Metabolism: Molecular Mechanisms Grant Objective: to identify factors affecting triglyceride synthesis and catabolism in enterocytes capable of regulating storage and secretion of triglyceride and with ultimate potential for regulating blood triglyceride concentrations. Translation: Identification of targets or methods for preventive and therapeutic interventions for obesity, CHD, and diabetes.
