ABSTRACT The in vitro toxicity test of water phase from ethanol extract of Justicia gendarussa Burm.f leaves to human normal lymphocyte cell According to previous research on antifertility activity of Justicia gendarussa Burm.f, it was found to be potentially developed as herbal medicine. Hence, it must be followed by premedical research then continued to clinical trials. One of the premedical research is toxicity test. Its need to be done to know the safety level usage. This research was intended to find toxicity effect of water phase from ethanol extract of Justicia gendarussa Burm.f leaves on lymphocyte cells culture by addition of MTT (3-(4,5-dimethilthiazol-2-yl)-2,5-diphenyl tetrazolium bromide). The consentration of test samples are 126 ppm, 252 ppm, 504 ppm, and 1008 ppm. 2x105 cells/mL suspension of lymphocyte cells were added in 96 tissue culture clusters. The water phase of Justicia gendarussa Burm.f leaves were added to the wells with that consentration in triplicate, then incubated for 24 hours. Following the addition of MTT, the cell was incubated further for four hours, and then the reaction was stopped by addition of SDS, and measured the optical density (OD) at wave length 550 nm using ELISA reader. The dead cells were then determined based on the OD. The result showed that water phase from ethanol extract of Justicia gendarussa Burm.f leaves have LC50 value 3215,7242 ppm.
