  1. 8 STEP GUIDE INTELLIGENT S3 ENVIRONMENTAL TOXICITY TEST IN 8 SIMPLE STEPS For use with the NG READER To ensure accurate results: please view the entire file to familiarise yourself with the protocol to enable fluidity of experimentation when carrying out the test.   2. STEP 1: Collect Test Sample Collect soil or other environmental sample for analysis using a clean trowel or equivalent. Take a sample of soil from the required depth ensuring each sample contains approximately 0.4kg of fine material/soil which excludes vegetation and stones/ pebbles which are greater than 2mm. This should be placed in a clean sample container for transportation or into the red top extraction agent, Quik-X, for the next stage.

  3. STEP 2: Prepare Sample • Transfer sufficient soil (or other environmental sample) • to the red top tube A until the sample plus Quik-X reaches • the marked line on the tube. • Mix tube A by vigorously shaking for 45 seconds. The tube should • be shaken whilst being held horizontally to maximise extraction • of the sample matrix. • Leave tube A to settle until between 1-2 cm of clear liquid • forms at top of the tube. The time taken for the sample to • settle will vary with different types of matrix and soil, so leave • the red top tube to stand whilst carrying out the next stages.

  4. STEP 3: Switch on Reader • Press and hold the red power button. • Allow the reader to perform self check analysis to ensure • accurate results, after this you will be presented with the • home screen menu.

  5. STEP 4: Activate Biosensor Whilst the sample is settling, prepare the biosensor bacteria (tube D) for use by adding the entire contents from tube C to tube D using one of the disposable pipettes. Resuspend the biosensor by sucking the liquid up and down 6 times. Discard pipette after use. Allow the biosensor to stand for exactly 3 minutes prior to use.

  6. STEP 5: Measure Base Level After the preparation period and using one of the syringes, draw biosensor suspension up to the marked line on the syringe and transfer to the test cell (tube E). Place the black HoldTite™ device into the top of the test cell tube and insert into the chamber of the S3 BioReader. Open the chamber by sliding the chamber release button to the left. The chamber lid is sprung and will open automatically once released. Close by pressing lid back down firmly. Press “Measure” and allow the instrument to collect the first reading (this takes approximately 10 seconds). The initial light result should be at least 10,000 RLU. If not, discard the test kit.

  7. STEP 6: Extract Test Sample Prepare the soil sample for analysis by transferring liquid from the cleared zone at the top of the red top tube A in the second syringe (fill to the mark) and add to tube B. Mix tube B by shaking vigorously for 15 seconds. The contents will become very frothy, this is quite normal and expected.

  8. STEP 7: Test Sample Open the chamber of the BioReader™ and remove the HoldTite™ plus test cell tube and separate the two. Using the remaining clean pipette, fill to the line and add to the test cell. Flick the tube to mix thoroughly till a light froth forms and replace the HoldTite™ plus test cell into the BioReader™. Using the four-way key pad, move the cursor onto the sample point for which you have already obtained a reading and press “Measure”. The instrument will read this second reading as a “Retest” and display this. For this sample point, you now have two readings, one initial light reading acting as a control from the biosensor light output and one measuring the effect of your sample on the biosensor.

  9. STEP 8: Analyse Result The % inhibition which is directly proportional to the toxicity of the test sample may now be calculated using the toxicity calculator app on either the supplied tablet or using a Smartphone or laptop / PC. The output from this app reports a sample as either “PASS” or “FAIL” depending on the two readings obtained. A “FAIL” indicates the soil contains levels of toxic contaminants above the action level and to be considered as contaminated.
