Build Day – Feb. 11 • Hands on soldering and project experience • Assemble electrosurgery tester kits from EWH National • Kits used in low resource settings to diagnose malfunctioning equipment • $5 to help cover costs • $20 for us to order
Duke Summer Program • EWH Summer Institute • Tanzania and Central America • One month training, one month work experience in developing hospitals • Learn the language, take technical classes, get lab experience, work in local hospitals • Applications due January 28 • http://ewh.org/index.php/programs/institutes
RNA Extraction Team Jessica Van Allen
Process • Lyse blood serum • Remove components which hinder PCR • Remove salts, proteins, and other cellular impurities
Challenges • Take a process with significant user input and make it almost completely automated • No contamination • Durable • Easy to Use • Inexpensive
Design • Fluid metering system replaces pipettes • Fluid flow system replaces centrifuges • Areas of focus for the quarter: • Pressure control system • Valve configuration • Electronic timing • Tubing
Quarter Goals • Design a pressure control method • Identify valve dimensions and configuration • Identify material and dimensions for tubing • Link system to electronic control
Thermocycler Team Justin Wong, Alexander Lin
Process • Obtain Sample from RNA team • Heat Sample & Cool Sample according to protocol • Export product to Gel team
Challenges • Temperature uniformity • Ramp rate (Heating/Cooling everything quickly) • Contamination • Cost • Durability/Repeatability
Quarter Goals • TEC (thermoelectric cooler) chip optimization • Rough working prototype
Gel Electrophoresis Team Kathryn Gent
Gel Electrophoresis Objectives • Qualitatively determine the presence of HIV in the blood • Based on open source designs, build a functioning gel box for electrophoresis
Process • Collect samples from Thermocycler team • Load samples into agarose gel • Run gel using electrophoresis • Analyze gel using DNA dye
Challenges • Building a gel box that will accurately run the gel, but less expensively than a pre-made box • Safer alternatives to ethidium bromide • Determining ideal power source based on electricity standards
