Detection of pMyBK1 ssDNA Intermediates via Southern Blot Hybridization

This study focuses on the detection of pMyBK1 ssDNA intermediates using Southern blot hybridization techniques. Total DNA from the Mycoplasma yeatsii type strain GIH.TS was analyzed on a 0.8% agarose gel, both with and without prior S1 nuclease treatment. The subsequent Southern blot, performed under non-denaturing conditions, utilized a digoxigenin-labeled pMyBK1 probe. The results highlight the presence of ssDNA intermediates, providing insights into the molecular characteristics of Mycoplasma yeatsii and its DNA stability.
