Collaborative Research: Mechanisms of Hybridization Kinetics in DNA Surface LayersRastislav Levicky, Polytechnic University, DMR-0706170 Surface DNA layers are extensively used in biomolecular diagnostics, from gene expression and genotyping to identification of forensic specimens and pathogen strains, yet many aspects of their physical behavior remain mysterious. This lack of fundamental understanding seriously impairs applications in clinical, research, and biothreat detection settings. Under “competitive” conditions typical of applications, thousands of solution nucleic acid strands with differing sequences compete for hybridization to a given surface-bound strand. This research seeks a detailed experimental and theoretical picture of competitive hybridization that is essential to effective experimental design and interpretation of these widely implemented, highly versatile diagnostic tools. Ferrocene tags (yellow star), conjugated to the sample strands, are used for quantitative electrochemical monitoring of the hybridization reaction. Top Right. The project is also providing the main thesis focus for graduate students Dr. Damion Irving and Dongbiao Ge, as well as lecture material for the upcoming course Biointerfacial Engineering that will be developed by the P.I.
