Notonly deadly infarct and stroke, but overall well-being. 3. 6 out of 10 deadliest risk factors directly relate to AS. 4. No. 1 “risk factor” of AS is diet and life-style. 5. AS is a degenerative disease.

  7. Why diet? Why physical inactivity? Physical inactivity Activity: beneficial effect on arterial stiffness Inactivity → obesity: dyslipidemia, metabolic syndrome, abdominal (visceral) fat as a dysfunctional “immune organ” that produces inflammatory cytokines Diet (in “developed” countries): Junk food (in every sense), Additives B-type malnutrition Waist circumference correlates stronger with CVD and MD than BMI.

Problems at the beginning… (chance to treat the initial cause?) • I/1 eNOS decreases with age • I/2 NO reacts with free radicals: • NO + O2- = ONOO- • I/3 ADMA that competes with L-arg • (Arginine-paradox) • II LDL-ch is VERY sensitive to oxidation • by free radicals • So, one thing is for sure: we need • efficient antioxidants. • And what else?

  12. Chance to treat the cause? An example: resveratrol Just look for the white arrows…

(reviewing the literature) 9 papers on FMD improvements by resveratrol (1921 papers all together on resveratrol benefits) 1 paper suggesting Q10 supplementation to decrease Aix 11 papers to show that grape polyphenols (antho- and procyanidines) improve FMD 3 papers on vitamin K2 decreasing vascular calcification 3 papers on plasma vitamin D decreasing arterial stiffness 21 papers to show that vitamin D improves other circulation parameters (coronary flow, for instance)

Our results… 2 x 1 daily placebo or 150 mg grape seed procyanidines + 60 mg Aroniamelanocarpaanthocyanidines; test at the beginning and after 3 months PWV (m/s) Treated group (n=11) 0 3 12.41±1.13 9.74±0.82* * significant at P < 0.05 • Also significant reduction in systolic BP • Some decrease in LDL-ch (n=7)

Our results… Regimen 1: 2 x 1 daily 150 mg GSE pro- + 60 mg Amanthocyanidines+ 10 mg resveratrol + 50 mg Q10 (water soluble) Regimen 2: regimen 1 + (2x1) 50 µg vitamin K2 and 1000 IU of vitamin D3) • Also significant reduction in BP • Significant increase in HDL-ch (n=5) • Significant improvement in EF
