Timeline • Thursday (3/27): Talk about how to devise a hypothesis, come up with a materials list, to-do list for the experiment, making out a schedule for the project, and go over lab report-meeting with me during the lab time, we will have a schedule to sign up • Friday (4/4): Hypothesis due, Materials list due, and schedule due- “Outline” due • Thursday (4/10): Round 1 of experiments. You will be keeping a lab notebook as usual. You come up with the title, purpose, methods, and results. • Thursday (4/17): Analyze results, put together notebook, and start writing lab report. • Monday (4/21): Lab notebook due • Monday (5/5): Lab report due.

  3. You Have Your Question…Now What? Devise A Hypothesis • What is a hypothesis? • An educated guess. But, it really isn’t a guess at all! • Do some internet research! Find out if this question has been looked at before. Or think about your question and make logical conclusions based on what you know! • Where do you look? • Credible resources: journals, textbooks, news articles, • What is not credible? Wikipedia, random internet sites that are not a credible news resources, and purely guessing

  4. Let’s Work Through An Example • Question: Which meat has the most bacteria? Oscar Mayer Hot Dogs or Oscar Mayer Sliced Turkey • Google Search: Bacteria in Lunch Meat • http://www.foodsafety.gov/poisoning/causes/bacteriaviruses/listeria/ • http://www.cdc.gov/pregnancy/infections-listeria.html • http://pittsburgh.cbslocal.com/2011/05/05/cold-lunch-meat-could-be-source-for-bacterial-infection/ • Conclusion: Most people get sick after eating cold cut meats.

  5. Hypothesis Cold cut sliced turkey from Oscar Mayer contains the most bacteria.

Another Example… • Question: Who’s hand in your group has the most bacteria? • This question is harder to find information for because it is very person-specific. • What do we do? • First, google search: Bacteria on hands • http://www.health.state.mn.us/handhygiene/why/handsbacteria.html • Now, what? Survey the group and see who washes their hands the most and least, see who touches the most surfaces or their face throughout the day, discuss your daily routines and where you go and the surfaces you touch.

  7. Hypothesis Dr. Crump has the most bacteria on her hands in the group.

  8. Now What? • Let’s test the hypothesis! • How? • Devise an experiment to test the hypothesis. • Detailed methods are the key!

  9. Let’s Work The Lunch Meat Example • Question: Which meat has the most bacteria? Oscar Mayer Hot Dogs or Oscar Mayer Sliced Turkey • What reagents do we need? FILL IT OUT ON YOUR MATERIALS LIST!! • Oscar Mayer Hot Dogs • Oscar Mayer Sliced Turkey • A way to cut or grind it up: scissors • 6 Nutrient agar plates • A way to dilute the ground up lunch meat: 6 microcentrifuge tubes, sterile water, sterile tips • Spreader • Ethanol • Bunsen Burner

  10. Methods • Grind up 1 gram of each lunch meat into 1 mL of sterile water, separately using scissors. Ethanol and flame the scissors in between each lunch meat to make them sterile. • Make serial dilutions: A: undiluted, B: 1:10, C: D: 1:100, E: 1:1000, F: 1:10,000 in sterile water and sterile microcentrifuge tubes • Plate 100 microliters of A-F dilutions with a sterile tip on a new nutrient agar plate. Be sure to ethanol the spreader, flame it, and let it cool prior to spreading • Place plates in the 37oC incubator upside down for 24 hours.

Make A Timeline… • Determines who does what. • Diagram it out: • 4/10-First experiment: • Dr. Crump: grinds the meat • Suzie: Adds sterile water to dilution tubes • Matt: Makes the dilutions with the lunch meat • Jessica: Plates the dilutions • 4/11-Suzie takes plates out of incubator and puts in refrigerator • 4/17-Analysis • Dr. Crump-Counts all the plates • Matt-Checks Dr. Crump’s counts • Suzie: Determines the number of bacteria • Jessica: Checks the total number of bacteria from Suzie • All work together to come to the conclusion (X had the most bacteria) or (the experiment didn’t work-why? What went wrong? Did we skip a step?)
