The therapeutic candidate inhibits FcR-triggered, Syk-dependent cytoskeletal rearrangement during phagocytosis. As stated by Rigel Pharmaceuticals, fostamatinib has a unique mechanism of action, blocking IgG receptor signaling in both macrophages and B cells via SYK kinase. The company is focusing on the fostamatinib ITP program specifically on the chronic form of this disease targeting the underlying autoimmune cause of the disease rather than stimulating platelet production. Moreover, fostamatinib is also being evaluated for autoimmune hemolytic anemia, IgA nephropathy, graft-versus-host disease, and ovarian cancer.
