  1. Synthesis and Characterization of Amphiphilic Dendron Coils – Potential Nanomicelle Precursors Jeromy T. Bentley University of Illinois at Chicago RET Fellow Department of Biopharmaceutical Sciences Advisor: Dr. Seungpyo Hong Mentor: Ryan Pearson July 28, 2011

  2. Summer Plan • Synthesize functionalized lipophilic polymer “coil” • Conjugate hydrophilic PEGylateddendron to lipophilic polymer coil to create amphiphilicdendron coil and induce self-assembled structure formation • Characterize synthesized amphiphilicdendron coils by 1H NMR • Characterize self-assembled micelles by TEM.

  3. Background - Dendron Coils • Dendron Coils – linear polymer that is dendronized at one end. Amphiphilicity can be engineered to allow for self-assembly. • Nanomicelles – very small self-assembled structure (<200 nm) capable of being used in drug delivery

  4. Dendron • Dendron – portion of a dendrimer (lots of branching) dendron dendrimer

  5. “Coil” Synthesis PLA-OH

  6. “Coil” Functionalization

  7. “Coil” Functionalization

  8. Conjugation of Dendron to Coil

  9. PEGylation of Dendron Coil

  10. Amphiphilic Dendron Coil dendron hydrophilic polyethylene glycol lipophilic polylactide coil

  11. Self Assembly to form Nanomicelle Nanomicelle: Lipophilic Core, Hydrophilic Periphery
