  1. Issues in Cytokine Measures: Talking points Debra Lyon RN FNP

  2. Absolute or Relative Multiplex More measures Approximation of a network? Substantial cost and sample savings ELISA Dynamic range (range over which there is a linear relationship between the cytokine concentration and the absorbance reading) is narrow.

  3. ELISA Gold-standard? More comparability Tried and true measure

  4. Multiplex • Flow cytometric multiplex arrays, also known as bead-based multiplex assays, • Employ proprietary bead sets which are distinguishable under flow cytometry. Each bead set is coated with a specific capture antibody, and fluorescence or streptavidin-labeled detection antibodies bind to the specific cytokine-capture antibody complex on the bead set.

  5. Sample Issues • Venipuncture • Plasma or serum? • Tubes? • Storage

  6. Problems for All • Low levels of circulating cytokines • Variability of cytokines over time • Can you compare across studies

  7. How to proceed? Conceptualization of multiplex and ELISA as “complementary tests, with the former being used more for discovery, while the latter may play a confirmatory role.” Xin Zhou, Maren S. Fragala, Janet E. 8. Bottom Line • Relationships among cytokines and sickness behaviors, symptoms and other phenomena of interest have resulted in better reproducibility in animal models than in human subjects.
