Germplasm Bank of Wild Species (GWOBS) passed national acceptance in Nov 2009 • 57618 accessions of seeds of >8141 species, and 7000 DNA samples by Dec 2012; • Good base for plant DNA barcoding • Voucher specimens for some seed collections with only vegetative characters; • Big challenges for species identification • The rapid, efficient and accurate species identification with DNA barcodes for these collections is one of the key tasks for GBOWS.

  7. Project: Plant DNA barcoding of China based on Germplasm Bank of Wild Species • Research period: July 2009-Dec. 2012 • Project number: 2009-LSF-GBOWS-01 • Participant: More than 50 botanists from 22 organizations Kick-off meeting for the project14th Aug., 2009

Main research contents • To construct standardized methods and protocols for DNA barcoding • To evaluate DNA barcodes for Plant DNA barcoding • To generated DNA barcodes for the seeds in GBOWS • To generate the standardized DNA barcodes for key and represented taxa • To construct DNA barcodes database system.

  9. Project task and objectives • First stage (2009-2012) • to generate 72 000 barcodes for 4 500 species, which include 3 600 DNA barcodes for 2 000- 3000 species from GBOWS . • Second stage (2013-2016) • to obtain 14 400- 150 000 DNA barcodes for 9 000 -10 000 spp, which will facility the rapid and accurate species identification for more than one third of Chinese vascular plants.

  10. Project progress • Prepare the methods and protocols for plant DNA barcoding • Two training workshops in January and August 2010 in Kunming Plant DNA barcoding technique training

  11. Jan. 5, 2010 Planning meetings • Discussed the developing strategy of plant DNA barcoding in China after the Mexico meeting. • Attained more DNA barcodes data in a short period; • Evaluation the four DNA barcodes; • To publish ‘high- impact’ papers.

DNA barcodes evaluation based on a large dataset • A total of 6286 individuals of 1757 species (1675 angiosperms and 82 gymnosperms) were used for universality analyses, and a total of 21673 sequences were obtained (5826 for rbcL,5471 for matK, 5566 for trnH-psbA, and 4810 for ITS); • ITS was suggested to be part of the core barcode for plants

  16. 13 articles of plant DNA barcoding published in this special issue; • All of them were supported by the grant from the Large-Scale Scientific Facilities of the Chinese Academy of Sciences (No. 2009-LSF-GBOWS-01).

  18. Barriers and threats • DNA barcoding is becoming a fashion in many areas in China but some do not understand the principles of DNA barcoding • China is a centralized country but with dicentralized funding and different funding agencies have different criteria • Lack of qualified taxonomists and many young students play DNA barcoding without a proper training in taxonomy • Problems in getting data from other parties
