  2. INTRODUCTION • Brand: Pioneer Quik Stripe Athletic Field Marking Paint • Aliphatic Hydrocarbon : toxic?? • Decreasing the amount of bacteria in the soil • Bacteria is helpful to soil • We want to discover if spray paint is actually harming our fields

  3. Procedure -Plot a soil area that is 2.5 meters long and 25 cm wide. -Mark flags on the ground, starting at 0 meters and mark after every .5 meter, there should be 6 flags. Label A, B, C, D, E, F.

  4. *10 cm away from each flag, place soil extractor in soil. *Go down 5 cm and place the soil into a labeled plastic bag. 1

  5. Cut one petrifilm sheet down the center (in half). Label 10^-3 and 10^-4 for each soil sample (A,B,C,D,E, F) Place 1 cc of soil sample into a culture tube containing 10 mL of sterile water; cap the tube and shake vigorously. Using a serological pipette, remove 1 mL of the soil/water mixture and place into a culture tube containing 9 mL of sterile water. 2

  6. -Repeat previous step using the second dilution tube and then repeat with third dilution tube. -Take 100 µl of the mixture 10^-3 and place it onto a Petrifilm sheet. -Repeat steps with mixture from 10^-3 to 10^-4 for all soil samples (A,B,C,D,E, F) 3

  7. Make cardboard template 10 cm by 5cm. Using template spray paint into rectangle 40 cm above ground for 5 seconds. Do this for plots A, C, E 4

  8. -Wait 5 min and pour 500 mL of water for each patch of soil -do plots B,D, F even though they were not spray painted -Take out 5 cm of soil using soil extractor for each plot -place in labeled plastic bags 5

  9. Do Bacteria Dilution process again for the new soil samples! 6

Conclusions • Aliphatic hydrocarbons would decrease the density of bacteria in the soil • Bacteria is reproducing and growing rapidly each day- such a high increase in the bacteria that was in the soil without spray paint • Hazardous not only to the soil, but also to the surrounding environment • Without the bacteria in the soil one of the major ways to get energy to plants would not be able to happen • Without the bacteria: • Nitrogen in the air would not be able to transform into ammonia (used for plant energy) > it would not provide energy for animals that eat those plants • Dead plants and animals don’t have nitrogen- they will not survive without nitrogen • Carbon cycle would not be able to complete > Carbon dioxide could not reach the plants > they would not be able to receive the glyceraldehyde phosphate > would not get the molecules: oxygen, nitrogen, hydrogen, carbon, and phosphate • Everything that is not received by the plants is not received by their consumers; animals. • The spray paint is included with a toxin that is killing the bacteria when it is sprayed on the soil.
