  1. Design • assume one baseline scan and one challenge scan • each scan starts with a bolus injection of [11C]raclopride

  2. Preliminary Results Baseline BP = 4.44 0 Baseline BP = 4.44 non-uniform sensitivity Baseline BP = 4.44 0

  3. Smoking three cigarettes over 90 minutes may cause diminishing dopamine responses

  4. Smoking three cigarettes over 90 minutes may cause diminishing dopamine responses

  5. Simulations suggest diminishing sensitivity over time.

  6. Dopamine Response to two cigarettes can be estimated at individual voxels (a.) Three voxel-based TACs from left dorsal caudate (DC) of smoker (grey dotted) are denoised by HYPR [1] to yield smoother TACs (red) which retain key temporal features. (b.) Fit of linearized ntPET [3] gives significantly improved fit (solid red) to data at single voxel in left DC, compared to standard MRTM model (dotted blue). (c.) Estimated DA responses of 10 voxels in left DC of same smoker all show similar timing. (d.) F-ratio map shows voxels with significant DA response; green arrow indicates cluster of voxels that pertain to plots (a-c). F-ratios in the cluster are much higher than the p<0.05 cutoff.
