  1. + Konjugacija / transpozicija : prenos transpozona Tn1731 z determinanto rezistence proti tetraciklinu iz seva RU4405 na plazmid F’ proAB lac v sev TOP10 CSH 100 F’ proAB lac RU4405 Tn1731 Tcr Iz plošče z gojiščem BHI zajamemo polno cepilno zanko zrasle kulture in razmažemo na ploščo z gojiščem LB in dodanim tetraciklinom. X gojišče BHI, 37oC preko noči Na takem selekcijskem gojišču zrastejo bakterije seva RU4405 Tn1731 Tcr ter RU4405 Tn1731 Tcr /F’ proAB lac Tn1731 Tcr Ploščo inkubiramo 2-3 dni pri 30oC, da se v nekaterih celicah seva RU4405 Tn1731 Tcr /F’ proAB lac transposzon Tn1731 prenese (transponira) na plazmid F’ proAB lac in dobimo sev RU4405Tn1731 Tcr/F’ proAB lac Tn1731 Tcr F’ proAB lac

  2. + Smr Smr Tn1731 Tcr F’ proAB lac Plazmid F’proAB lac Tn1731 Tcr nato prenesemo v sev TOP10 Smr tako, da nastavimo naslednjo konjugacijo. Na ploščo z gojiščem BHI razmažemo polno cepilno zanko bakterij zraslih na plošči z gojiščem LB in dodanim tetraciklinom in sev TOP10. Razmazane kolonije inkubiramo preko noči pri37 oC. Transkonjugante izoliramo tako, da zajamemo polno cepilno zanko zraslih bakterijskih celic in jih razmažemo na ploščo selekcijskega gojišča,ki je v tem primeru LB z dodanim tetraciklinom in streptomicinom. X Tn1731 TOP10 F’ proAB lac Tn1731 Tcr Top10 Smr / F’proAB lac Tn1731 Tcr Nato razmažemo zrasle kolonije na ploščo s selekcijskim gojiščem: LB + Tc + Sm in Inkubiramo preko noči pri 37oC Inkubiramo preko noči pri 37oC
