  1. Extracellular nucleic acids from blood and their using for noninvasive diagnostics and monitoring of cancer PhDTamkovich S.N. GrantY4-B-08-13 REC-008 Novosibirsk State University Institute of Chemical Biology and Fundamental Medicine SB RAS Intrainstitute Center of DNA Sequencing SB RAS The objective of the research istheinvestigation of features of tumor-specific gene alterations circulation on the whole extracellular nucleic acids and search of new DNA-markers in blood cancer patients plasma and cell-surface-bound extracellular nucleic acids.

  2. The characterization of circulating DNA from blood plasma of healthy donors and breast cancer patients The experiment strategy: The isolation of plasma DNA from blood The hydrolysis of plasma DNA by EcoRI и Bsp143I The ligation of plasma DNA into pBlueScript II KS(-) as vector:insert= 500 ng : 50 ng The transformation of XL-Blue E. coli by electroporation The selection of white colonies and screening by PCR The production and isolation of plasmid recombinant DNA The sequencing and analysis of cloned DNA fragmentsby BLAST server (http://www.ncbi.nlm.nih.gov/BLAST/)

  3. Chromosome localization of circulating DNA Healthy female Breast cancer patients

  4. Sequence analysis Breast cancer patients Healthy female • Identified sequences in blood of breast cancer patients: • Repetitive elements - • Alu, LTR, (CAG)n, L1, THE, MST, MIR • Genes, associated with breast cancer - • GSTA1, ANKRD6, ZNF217, DKFZp566C0424

  5. RESULT: Sequence analysis revealed that the number of circulating DNA fragments does not depend from the size of paternal chromosome both in normal and pathological state. Fragments from chromosome six were found in the healthy female bloodstream more frequently. BLAST analysis demonstrates that 10% of coding DNA fragments circulating in blood of the breast cancer patients is associated with development of this pathology. Moreover, in breast cancer patients blood frequency of genes, pseudogenes and CpG islands was below that in healthy female blood. The unidentified sequences of human genome and repetitive elements in health are presented loss frequently then in disease.
