  1. PERKIN-ELMER MODEL 200 UV-VIS SPECTROPHOTOMETER OPERATING INSTRUCTIONS

  2. INSTRUMENT WARM UP • Turn the power switch on. Turn on the tungsten lamp (W lamp). The tungsten lamp is used up to 370 nm. Allow the lamp to warm up for a few minutes. (30 minutes are necessary for stable operation). • Set the source selector to the Visible lamp (out). • Set the slit to the minimum, 0.2 nm. This setting gives sufficient resolution for recording spectra of gas-phase samples. Set the response to MEDIUM for now.

  3. %T and Absorbance Calibration • Depress the %T button. Insert the 0%T shutter (large plate) along the side of the sample compartment, blocking the sample beam.

  4. %T and Absorbance Calibration •  Set +00.0 on the digital display using the 0%T control. Turn control enough to see –00.0 then turn back until +00.0 just appears. • Remove the 0%T shutter. Set +100.0 with the 100%T/ ABS. Control.

  5. Absorbance Spectrum • Push the 0-2 Abs button to switch to Absorbance readout. • Set spectrophotometer SCAN SPEED selector to 30 nm/min. The drive gears should be arranged as shown below.

  6. Absorbance Spectrum • Set the RESPONSE switch to FAST, to follow the sharp gas-phase peaks. • Place the sample cell in the sample compartment.

  7. Absorbance Spectrum • Manually set the initial, longest wavelength, in the monochromator. • When setting wavelengths, always turn the WAVELENGTH control from longer to shorter wavelengths to avoid backlash in the gears for best reproducibility. • Click the Start Green button on LoggerPro • Switch the Scan Box switch to ON • After data collection click the red Stop button in Logger Pro and turn off the Scan Box switch
