  1. Naegleria gruberiaA Model Organism for Undergraduate Learning 1. Normally an amobea, however under conditions of stress rapidly (120 min) transform to a flagellated form. 2. This transformation requires gene transcription. 3. Publically available microarray studies have been preformed. (Fritz-Laylin and Zacheus Cande, 2010) 4. Genome has been sequenced.

  2. Naegleria Transformation Time Course

  3. Target Audience and Timeframe Cell and Molecular Biology Laboratory One semester 3 hour lab Project could be expanded over several semesters and involve many students.

  4. Objectives • Internalize the concept of gene expression changes controlling cell structure and function • Develop skills in microscopy and imaging • Understand the basic principles of microarray analysis • Develop skills in data manipulation and analysis • Develop skills in scientific writing and data presentation • Introduce the use of PCR in cloning

  5. Projects/Activities Growth of Naegleriaand transformation to flagellated form. Imaging of Naegleria Develop PCR primers and clone one or more genes required for transformation. Data mine genome and microarray data for additional genes involved in transformation.

  6. Assessment Mechanisms Problem based essay question. Poster presentation
