  1. GK-12 Saturday Workshop RS: Roxanna Ochoa CT: ChariePoderoso October 20, 2012

  2. In this lab, students will be introduced to restriction enzymes, DNA profiling and gel electrophoresis

  3. Standards Addressed • Cell Biology 1a, 1c • Genetics 2d, 2e • Investigation & Experimentation 7a, 7c

  4. Timeline Day 1 Day 2 DNA fingerprinting Worksheet DNA fingerprinting video Students will observe gel and determine guilty suspect • Introduce DNA fingerprinting • RE and electrophoresis activity

  5. DNA • DNA makes you unique. Your DNA is different from every other person in the world. • An individual’s DNA forms a unique pattern of bands that can be used to identify the person. • This unique banding pattern produced by fragments of your DNA is called DNA fingerprinting. • Law enforcement is able to gather the smallest amount of evidence at a crime scene and compare it to DNA from a suspect’s blood or saliva.

  6. Day 1: Restriction Enzymes and electrophoresis • Introduce restriction enzymes and electrophoresis (PPT presentation) • Activity (Restriction Enzymes and electrophoresis Worksheet)

  7. Restriction Enzymes and electrophoresis

  8. Day 2: Crime Scene • Introduce students to crime scene scenario • Class demonstration • Load gels with suspect and crime scene DNA (FisherScientific #S97676) • Simplify lab by using dye samples (kit or make own) • During gel electrophoresis:DNA fingerprinting video. • Analyze data

  9. Lab Worksheet

  10. Alternative • NOVA Fingerprinting

  11. High School Modification • Students can make their own gels and prepare their own DNA • Explain electrophoresis and restriction enzymes in more detail • Change crime scene scenario
