  1. Denaturing Urea PolyacrylamideGel Electrophoresis

  2. 15% Native PAGE-TBE gel • 1M, 2M, 3M Urea gels • 36 base pair DNA

  3. 15% Native Observations: Samples show some excess Cy5 or Cy3. Samples Ligated, Nicked AT, Nicked GC show the same mobility .

  4. 1 M Urea Observations: There was no difference in mobility between the Ligated, Nicked AT, Nicked GC.

  5. 2 M Urea Observations: There was no difference in mobility between the Ligated, Nicked AT, Nicked GC.

  6. 3 M Urea Observations: Samples were denatured in the gel. Could not make any conclusions about mobility.

  7. Conclusion: The 3M Urea gel denatured the samples and mobility could not be determined. The 15% native, 1M Urea, and 2M Urea gels did not show difference in mobility.
