  2. Before you start get or make a copy of this Table of results

  3. Step 1 Collect leaves. Tie back hair and wash hands.

  4. Step 2 Wash and disinfect the bench. Flame the cork borer to sterilise.

  5. Step 3 Place two agar plates on the bench. Label ‘A’ and ‘B’.

  6. Step 4 Cut as many discs as possible from the leaf using the cork borer.

  7. Step 5 Place plate ‘A’ upside-down on the bench.

  8. Step 6 Flame the forceps and allow it to cool.

  9. Step 7 Carefully raise the base of the petri dish just enough to work with.

  10. Step 8 Using the forceps, smear dots of petroleum jelly onto the inside of the lid.

  11. Step 9 Use the forceps to attach a leaf disc to each blob of jelly. Re-flame the forceps.

  12. Step 10 Seal the plate with parafilm. Seal plate ‘B’ with parafilm. Label and date both plates.

  13. Step 11 Store all petri dishes right way up in an incubator at 20°C for 24 hours.

  14. Step 12 Turn the petri dishes upside down and incubate at 20°C for 3 days.
