  1. Figure 2S – chkA and chkB mRNA expression increases during sexual development. Fold increase in chkA (A) and chkB (B) mRNA levels during sexual. Sexually developing cultures were initially propagated as vegetative cultures in liquid MM + CA medium (Han DM, Han YJ, Lee YH, Jahng KY, Jahng SH, Chae KS (1990) Inhibitory conditions of asexual development and their application for the screening of mutants defective in sexual development. Plates were sealed with parafilm and wrapped with aluminium foil in order to induce fruiting body formation specifically. Real-time RTPCR was used to quantify the mRNA. The measured quantity of the chkA and chkB mRNA in each of the treated samples was normalized using the CT values obtained for the tubC RNA amplifications run in the same plate.The relative quantitation of chkA and chkB and tubulin gene expression was determined by a standard curve (i.e., Ct –values plotted against logarithm of the DNA copy number). Results of four sets of experiments were combined for each determination; means are shown. The values represent the number of times the genes are expressed compared to the wild type control grown without any drug (represented absolutely as 1.00).
