Fig. 2: Effects of treatments on cell proliferation.SMC and M07e cells were cultured in 96-well plates and 100 µM db-cAMP (+ 100 µM IBMX), 300 µM 8-CPT or db-cAMP + IBMX in the presence of the inhibitors KT5720 (10 µM) or PD98059 (25 µM) as well as BrdU(10 µM)was added for 24 h.The plates were washed, and a chemiluminescentBrdU cell proliferation assay was performed according to the manufacturer’s instructions (Roche). Similarly to the results in M07e cells, a slight decrease in cell proliferation after addition of db-cAMP was observed in HeLa cells (not shown). (mean values ± SD; un-stimulated control =100%; n= 5).
