Factor Xa inhibitors FXa may be a better target than thrombin • Has few functions outside coagulation (compared with thrombin) • Has a wider therapeutic window than thrombin (separation of efficacy and bleeding), in vitro • Thrombin inhibitors are associated with rebound thrombin generation – no evidence with FXa inhibitors • Efficacy of heparin-based anticoagulants improves as selectivity for FXa increases: UFH < LMWH < fondaparinux
