Management of material in MV’s Molecular Laboratory: • MV’s tissue collection was started a couple of decades ago to essentially support the research of a few researchers. • The collection now includes tissue samples and genetic products which support the research of many research staff and their students. • In order to better manage this collection, a number of collection management and risk management policies and procedures have been developed that will: • Allow us to better manage and track the use of the collection • Allow us to database, track and manage all genetic products that accumulate from research in the Lab. – e.g. In recent years, students have undertaken such projects as researching the genetics of limb development in dragon lizards, and studying immune genes in a number of Australian bird species. • Record research using ancient DNA techniques on collection specimens that have been destructively sampled .

  3. MV’s new EMu Transactions Module is being used to record: • The use of MV tissue samples, including: • Tissue grants to external researchers • Tissues used for MV research • The destructive sampling of collection specimens – for both external and internal research • Incoming tissue grants to MV researchers and students. Incoming tissue grants: • Tissues are catalogued on EMu – recorded as ‘not part of the State Collections’ as they may remain the property of the granting institution. • Genetic products are catalogued on Emu – again recorded as ‘not part of the State Collections’. • Research on such tissues is recorded on the Emu Catalogue Tissue and Genetics Tabs.

  4. Transaction Record Gibber Earless Dragon Tympanocryptis intima All destructive sampling of MV collection specimens is recorded on MV Transactions module. Muscle tissue was removed from the hind limb of 3 paratypes for ancient DNA research.

  5. Catalogue Transactions Tab The use of the Transactions Module to record all destructive sampling and tissue grants ensures that the history of such use displays on the Catalogue record of the voucher specimen. Comments are mirrored from the Transactions Module

  7. Catalogue Parts Tab Displays the hierarchical relationship between the voucher specimen and the DNA extraction.

  8. Catalogue Genetics 1 Tab Records whether or not DNA was successfully extracted from the tissue – negative results are informative.

  9. Records: • The gene region that was amplified and sequenced, and the primers used. • The GenBank Accession number (hyperlink to GenBank record yet to come) • It is also important and informative to record non-results – ie. when researchers were unable to amplify DNA. • PDF of MPE publication is attached to the MMR Catalogue Genetics 2 Tab

  14. Destructive sampling of MV frogs Growling Grass Frog Litoria raniformis Tissues from specimens collected in the 1960’s are used to study human impacts on frog populations around Melbourne.
