A 276-bp fragment from the 3’-UTR of the IGF1R gene [position 7640 to 7916, NM_000875, containing two miRNA-7 binding sites (31)] and a 62-bp fragment from the 3’-UTR of RhoC gene [position 1210 to 1271, NM_175744, containing a known miR-138 binding site (13)] were cloned into the Xba I site of the pGL3 firefly luciferase reporter vector (Promega). Cells were transfected with these reporter constructs (named pGL-IGF1R and pGL-RhoC, respectively) and pRL-TK vector (as internal control for transfection efficiency) using lipofectamine 2000 (Invitrogen), and treated with miR-138 mimic, miR-7 mimic, or control mimic. Dual luciferase reporter assays were performed as described in the Materials and Methods section. * indicates p < 0.05.
