  1. DETERMINATION OF POP’S IN HUMAN SERUM SAMPLES OF ADOLESCENTS FROM THE FLEMISH ENVIRONMENT AND HEALTH STUDY (FLEHS II) Croes K., Den Hond E., Bruckers L., Colles A., Koppen G., Loots I., Nelen V., Schoeters G., Nawrot T., Van Larebeke N., Covaci A., Van Langenhove K., Vandermarken T., Elskens M. and Baeyens W. Department of Analytical and Environmental Chemistry

  2. FLEHS II 2007-2011 • Flemish Environment & Health Study – second cycle • Reference campaign (newborns-adolescents-adults) • 2 hotspots (Genk-Zuid & Menen): adolescents • More than 40 biomarkers of exposure and 10 effect markers • Information from questionnaires (asthma & allergy, lifestyle, food intake) POPs in human serum - Croes Kim

  3. Chlorinated POPs in adolescents • 14-15 years old adolescents (n=3x200) • Marker PCBs (138,153,180), PCDD/Fs, dl-PCBs, DDE & HCB in serum • Thyroid hormones, sexual development, asthma & allergy, lifestyle factors (boys & girls) • Sex hormones (only boys) POPs in human serum - Croes Kim

  4. Goals To generate reference values for Flanders for all biomarkers Compare hotspots to reference mean Compare results with (inter)national studies Establish dose-effect relationships POPs in human serum - Croes Kim

  5. Results: Chlorinated POPs • use of local food: less frequently local eggs, vegetables & fruit (p<0.001) • lower consumption of fat-rich food in general • time trend:hotspots =>2 years later POPs in human serum - Croes Kim

  6. Results: Corrected data • Stepwise multiple regression • Fixed confounders: BMI, sex, age, smoking, amount of blood lipids POPs in human serum - Croes Kim

  7. Time trends Corrected geometric means compared to FLEHS I (5 years earlier) • Sum of PCBs: 23% • p,p’-DDE: 26% • HCB: 60% • PCDD/Fs • Dl-PCBs POPs in human serum - Croes Kim

  8. FLEHS II compared to other human biomonitoring studies • FLEHS II (14-15 y, this study, 2008-2009); FLEHS I (14-15 y, 2003-2004) • US NHANES 4th report (12-19 y, ’03-’04); GerES IV (12-14 y, ’03-’06) • Sum PCBs - 282 ng L-1 or 66 ng g-1 lipids(FLEHS I) - 218 ng L-1 or 50 ng g-1 lipids (FLEHS II) - 266 ng L-1 (GerES IV) - 12.9 ng g-1 lipids (NHANES) • p,p’-DDE - 418 ng L-1 or 94 ng g-1 lipids(FLEHS I) - 309 ng L-1 or 70 ng g-1 lipids (FLEHS II) - 190 ng L-1(GerES IV) - 105 ng g-1 lipids(NHANES) • HCB - 91 ng L-1 or 21 ng g-1 lipids (FLEHS I) - 36.7 ng L-1 or 8.2 ng g-1 lipids (FLEHS II) - 91 ng L-1 (GerES IV) - 13.3 ng g-1 lipids (NHANES) http://www.cdc.gov/exposurereport/pdf/FourthReport.pdf http://www.umweltdaten.de/publikationen/fpdf-l/3355.pdf POPs in human serum - Croes Kim

  9. Dose-effect relationships • Sex hormones (boys) • - neg ass: (free) estradiol • - pos ass: (free) testosterone (except PCDD/Fs: neg) • Thyroid hormones • - pos ass: free T4 POPs in human serum - Croes Kim

  10. Dose-effect relationships • Sexual maturation (boys) • - pos ass: pubic hair & genital development • Sexual maturation (girls) • - neg ass: menarche, breast & pubic hair development POPs in human serum - Croes Kim

  11. Dose-effect relationships • Asthma & allergy • Pos ass: doctor diagnosed asthma • Pos ass: dl-PCBs & PCDD/Fs and animal allergy • Neg ass: dl-PCBs and eczema POPs in human serum - Croes Kim

  12. Conclusions • Chlorinated POPs lower than in FLEHS I • Follow up in time (FLEHS III): decrease? • Hotspots: sensitization seems important • Dose-effect relationships: • Limited literature data on adolescents • Difficult to interpret our results • Some date in agreement with FLEHS I (e.g. pos ass. with testosterone) • Others: opposite (e.g. neg ass with estradiol) • But still: stimulation for legal actions POPs in human serum - Croes Kim

  13. Acknowledgements This study was commissioned, financed and steered by the Ministry of the Flemish Community (Department of Economics, Science and Innovation; Flemish Agency for Care and Health; and Department of Environment, Nature and Energy). POPs in human serum - Croes Kim
