ABSTRACT THE INHIBITION OF APIGENIN AND HESPERIDIN TO HYALURONIDASE EC 3.2.1.35 ACTIVITY IN VITRO Flavonoid and its derivatives such as apigenin and hesperidin were reported to inhibit hyaluronidase activity. However, the inhibition types of apigenin and hesperidin to hyaluronidase activity is still unknown. In this study, the inhibitory effects of apigenin and hesperidin to hyaluronidase were measured using Morgan-Elson assay. The inhibition types of these compounds were determined by comparing KM and V0 max value of hyaluronidase with and without apigenin or hesperidin. These comparison were shown by Lineweaver-Burk curves. The results showed that apigenin and hesperidin at concentrations 0,25 to 1,00 mM inhibit hyaluronidase activity. The hyaluronidase activity was decreased to 76,30% after treatment with apigenin and 73,56% with hesperidin. The KM and V0 max value of hyaluronidase without inhibitor was 3,63 mM and 2,41 x 10-3 mM/minute. After treatment with apigenin, the KM and V0 max value of hyaluronidase was 12,19 mM and 2,21 x 10-3 mM/minute. Besides, after treatment with hesperidin, KM and V0 max value of hyaluronidase was 5,73 mM and 2,73 x 10-3 mM/minute. Based on this results, it is known that apigenin and hesperidin inhibit hyaluronidase activity by competitive-reversible inhibition. Keywords : apigenin, hesperidin, hyaluronidase EC 3.2.1.35
