What is genetics ? • “Genetics is the study of heredity, the process in which a parent passes certain genes onto their offspring.” • Offspring inherit their biological parents’ genes that express specific traits, such as some physical characteristics, natural talents, and genetic disorders.

Genetic concepts • Heredity describes how some traits are passed from parents to their offspring. • The traits are expressed by genes, which are small sections of DNA that are coded for specific traits. • Genes are found on chromosomes. • Humans have two sets of 23 chromosomes—one set from each parent.
Mendelian Inheritance • The inherited traits are determined by genes that are passed from parents to offspring. • A offspring inherits two sets of genes one from each parent. • A trait may not be observable, but its gene can be passed to the next generation.
Classical (Mendelian) Genetics • Principles of genetics were developed in the mid 19th century by Gregor Mendel an Austrian Monk • Developed these principles without ANY scientific equipment - only his mind. • Experimented with pea plants, by crossing various strains and observing the characteristics of their offspring. • Studied the following characteristics: • Pea color (Green, yellow) • Pea shape (round, wrinkled) • Flower color (purple, white) • Plant height (tall, short) • Made the following observations (example given is pea shape) • When he crossed a round pea and wrinkled pea, the offspring (F1 gen.) always had round peas. • When he crossed these F1 plants, however, he would get offspring which produced round and wrinkled peas in a 3:1 ratio.
Laws of Inheritance • Law of Segregation: When gametes (sperm egg etc…) are formed each gamete will receive one allele or the other. • Law of independent assortment: Two or more alleles will separate independently of each other when gametes are formed

Six basic genotype crosses เมื่อผสมพันธุ์สัตว์ที่มี genotype homozygous x homozygous ได้ลูกที่มี genotype แบบเดียว homozygous x heterozygousได้ลูกที่มี genotype 2 แบบ ½ : ½ heterozygous x heterozygousได้ลูกที่มี genotype ครบทุกแบบ
Law of independent assortment (กฎการรวมตัวอย่างอิสระของยีน) ยีนแต่ละตำแหน่งจะสามารถแยกไปกับอีกยีนหนึ่งได้อย่างอิสระเมื่อมีการสร้างเซลลสืบพันธุ์ และมีโอกาสมาเข้าคู่กันได้ใหม่อย่างอิสระด้วยเมื่อมีการผสมพันธุ์เป็น zygote กล่าวคือ ยีนแต่ละตำแหน่งจะสามารถเกิดกฎการแยกตัว (Law of segregation) ได้อย่างอิสระนั่นเอง กฎนี้กล่าวถึงยีนตั้งแต่ 2 คู่หรือ 2 ตำแหน่ง และควบคุมตั้งแต่ 2 ลักษณะขึ้นไป โดยแต่ละลักษณะนั้นแสดงออกอย่างอิสระต่อกัน ชื่อเรียกอื่น : • Mendel’s 2nd Law • 2nd Law of Genetics • Law of Random Assortment of Characters
Law of independent assortment (กฎการรวมตัวอย่างอิสระของยีน) Dihybrid Cross การผสมพันธุ์ระหว่างคู่ heterozygous genotype ของลักษณะที่ถูกควบคุมด้วยยีน 2 คู่ ในการคำนวณหา genotype และ phenotype ที่เกิดจากการผสมของยีนหลายตำแหน่ง สามารถทำได้ 3 วิธี ได้แก่ • Punnett Square Method • Algebraic Method • Branch or Forked-line Method
Sex and Color inheritance 1. Sex-Linked Traits ยีนมีตำแหน่งอยู่บนโครโมโซมเพศโดยตรง ดังนั้นการกำหนดเพศของสัตว์ (sex determination) จึงมีผลต่อการแสดงออกของยีน

Sex and Color inheritance 2. Sex-Influenced Traits ยีนมีตำแหน่งอยู่บนออโตโซม หากสัตว์มี genotype แบบ dominant หรือ recessive homozygous ก็จะแสดงออกเหมือนกันทั้งสองเพศ แต่จะควบคุมให้มีการแสดงออกในเพศแตกต่างกัน หากสัตว์มี genotype เป็นแบบ heterozygous

Sex and Color inheritance 3. Sex-LimittedTraits ยีนมีตำแหน่งอยู่บนออโตโซม แต่จะสามารถควบคุมให้มีการแสดงออกได้เฉพาะในเพศใดเพศหนึ่งเท่านั้น

Crossing over เป็นกระบวนการแลกเปลี่ยนชิ้นส่วนของยีนระหว่างโครโมโซม (non-sister chromosome)ที่เป็นคู่กัน ผลจากการเกิด crossing over จะทำให้เกิดการแลกเปลี่ยนชิ้นส่วนโครโมโซม (ยีน) ....... อาจส่งผลให้ยีนที่อยู่บนโครโมโซมเดียวกันไม่ถูกถ่ายทอดไปด้วยกัน .......
การเกิด crossing over … เมื่อสิ้นสุดการแบ่งเซลล์ไมโอซิส จะได้เซลล์สืบพันธุ์ที่แตกต่างกัน 2 ชนิด 1. Parental type (gameteชนิดที่ไม่เกี่ยวข้องกับการเกิด crossing over) 2. Recombination type (gameteชนิดที่เกิดจากโครมาติดเกิด crossing over)
แผนที่ยีน • แผนที่ยีน (genetic map) หมายถึงคือการนำยีนที่อยู่บนโครโมโซมเดียวกันมาเรียงลำดับ และเพื่อบอกระยะห่างระหว่างยีน บนโครโมโซม • ระยะทางระหว่างยีน (genetic distance) คำนวณได้จากอัตราการเกิด crossing over โดยกำหนดให้ 1% crossing over มีค่าเท่ากับระยะทาง 1 map unit หรือ 1centimorgan(cM)
ประโยชน์ของการทำแผนที่ยีนบนโครโมโซมประโยชน์ของการทำแผนที่ยีนบนโครโมโซม 1. ช่วยในการหาลำดับของยีนที่เรียงตามยาวบนโครโมโซม 2. สามารถหาระยะทางระหว่างยีนบนโครโมโซม (ระยะทางของยีนบนโครโมโซมจะวัดในหน่วยของ map unit) ใช้ในการศึกษาการแสดงของลักษณะต่างๆ ได้ทั้งในโครโมโซมของ prokaryote และ eukaryote 3. มีประโยชน์ในการศึกษาทาง molecular biology และ genetic engineering 4. เมื่อมีการผ่าเหล่า (mutation) ทำให้ทราบและบอกชนิดของการเกิดการผ่าเหล่าได้ทันที
Linkage gene ต่อการปรับปรุงพันธุ์สัตว์ • ประยุกต์ใช้กลุ่มยีนที่มีความเกี่ยวข้องกันและถ่ายทอดไปด้วย (linked)เพื่อเป็นเครื่องหมายทางพันธุกรรมช่วยในการคัดเลือกพันธุ์สัตว์ได้ • โดยเฉพาะลักษณะทางเศรษฐกิจที่ควบคุมด้วยยีนหลายคู่ .. การคัดเลือกยีนใดยีนหนึ่งเพื่อบอกบ่งชี้ทำได้ยาก .. เช่น ยีนควบคุมการให้ผลผลิตน้ำนม และยีนอื่นๆที่ควบคุมลักษณะเชิงปริมาณ เป็นต้น
IA, I B, I O Multiple allele : การควบคุมการแสดงออกยีน ณ locus หนึ่งประกอบด้วยยีนที่มี allele มากกว่า 2 อัลลีล ขึ้นไป เช่น กรุ๊ปเลือด ระบบ ABO IA,I B,I O IAIA, I AIO = Type A IBIB, IBIO = Type B IAIB= Type AB IOIO = Type O
Multiple allele • การควบคุมการแสดงออกโดยยีนมากกว่า 2 อัลลีนอยู่บนตำแหน่งเดียวกันบนคู่ โฮโมโลกัสโครโมโซม การมีสีขนในกระต่าย การมีสีขนในกระต่ายถูกควบคุมด้วยยีนคู่เดียวแบบ multiple alleles โดย C = Full color (ขนสีดำหรือน้ำตาลตลอดทั้งตัว) cch = Chinchilla (ขนสีเทาและขาวแซมตลอดตัว) ch = Himalayan (ขนสีขาวแต่มีสีดำที่ปลายหู จมูก หาง และเท้า) c = Albino (ขนสีขาว ตาแดง)
Pleiotropic gene • การที่ยีนเพียงตำแหน่งเดียวสามารถควบคุมการแสดงออกได้มากกว่า 1 ลักษณะ
Modifying Gene • Modifying gene คือยีนหรือกลุ่มยีนที่ทำหน้าที่ดัดแปลงการแสดงออกของยีนหลัก (major gene) โดย modifying gene ส่วนใหญ่มักมีอยู่หลายตำแหน่ง • Modifying gene โดยมากควบคุมในลักษณะปริมาณ ในขณะที่ major gene ที่เกี่ยวข้องมักเป็นยีนที่ควบคุมการมีลักษณะเชิงคุณภาพ
Modifying Gene Holstein Friesian • Ex. การมีสีขาว-ดำของโคนมพันธุ์ Holstein Friesian พบว่า การมีจุดขาวบนพื้นลำตัวสีดำนั้นเป็นลักษณะเด่น ซึ่งถูกควบคุมโดย major gene ในขณะที่ขนาดและตำแหน่งบนลำตัวของจุดขาวเหล่านั้นจะถูกควบคุมด้วย modifying gene อีกหลายตำแหน่ง

Lethal Gene • การควบคุมการแสดงออกโดยยีนโดยก่อให้เกิดลักษณะผิดปกติและอาจก่อให้เกิดการตายในที่สุด ลักษณะการตายในสัตว์
Lethal Gene • Dominant Lethal การควบคุมลักษณะของสัตว์ในลักษณะที่ dominant homozygous genotype จะตาย • Recessive Lethal การควบคุมลักษณะของสัตว์ในลักษณะที่ recessive homozygous genotype จะตาย • Semilethal สัตว์มีลักษณะผิดปกติเนื่องจาก dominant หรือ recessive lethal แต่ยังสามารถเลี้ยงให้มีชีวิตรอดได้ถ้ามีการจัดการดูแลอย่างเหมาะสม
