Nouveaux antirétroviraux Tolérance rénale du QUAD (1) CROI 2013 – D'après Ramanathan S. et al., abstract 524 actualisé • Étude randomisée et contrôlée de phase I réaliséechez 72 volontaires sains non infectés par le VIH et à fonction rénale normale (DFGe ≥ 90 ml/mn) : • Fonction rénale évaluée par : • DFGa (actuel, clairance au iohexol) – DFGe (estimé) • Flux sanguin rénal (clairance au para-aminohippurate – PAH) • Marqueurs de la fonction tubulaire (aminoacidurie, fraction excrétéede l’acide urique, bêta2-microglobinurie, ratio albumine urinaire/créatinine) • 5 bras (14 patients par bras) : COBI, COBI + TDF, STB, TDF, Placebo • Période sous traitement (30 jours) puis période de suivi (30 jours)

Nouveaux antirétroviraux Tolérance rénale du QUAD (2) CROI 2013 – D'après Ramanathan S. et al., abstract 524 actualisé • Résultats : 94,0 (89,3-99,0) • L’administration de COBI, TDF, COBI+TDF ou de STB chez le volontaire sain n’a pas d’impact sur les différents paramètres rénaux étudiés • Une légère diminution réversible du DFGe a été constatée sous COBIversus placebo, en rapport avec l’inhibition de la sécrétion de la créatininevia l’inhibition du transporteur rénal MATE-1
