  1. Luminex, Robotics & All That Stuff Erin Longdo VP Business Development Applied Cytometry Systems

  2. Overview • Introduction • Case Study – GSK HTB Harlow, UK • Future Directions – Applications • Future Directions – Software & Information Management

  3. GSK High Throughput Biology • Describe your group and goals/challenges here – copy and paste if you need more slides

  4. Process Analysis & Automation • Process Analysis & Automation supply quality control software to the pharmaceutical & petrochemical industries • PAA supply OVERLORD™ for the integrated control of laboratory automation workcells • PAA have recently developed OVERLORD™ Scheduler for time critical automation procedures • OVERLORD™ is used in ~200 workcells

  5. OVERLORD™ • OVERLORD™ was developed to control the National DNA database at the Forensic Science Service (FSS, UK) • OVERLORD™ is the link between the FSS LIMS and also controls all the automation equipment • This automation system has processed ~2m samples in the last 6 years.

  6. Collaborations • GSK High Throughput Biology Project Leaders • Goal = Improve Lab Efficiency & Proof of Principle for ????? • Process Analysis & Automation (PAA) • Integration of all systems in the workflow • Applied Cytometry Systems (ACS) • Integration Module for OVERLORD™ • Downstream data analysis & management

  7. Project Goals • Increase Lab Efficiency • Free up BRAIN POWER • Promote Access to Luminex Technology

  8. Phased Project • Phase 0 Luminex assay development & method validation. All Manual – up to 4 plates/day • Phase 1 Real-time integration of system using OVERLORD™ software – up to 8 additional plates/day • Phase 2 Integration using OVERLORD™ Scheduler (pre-emptive) – up to 20 plates/day

  9. System Components • OVERLORD™ Software • Tecan Genesis with Gemini software • Incubators (onboard the Tecan) • Plate washer (onboard the Tecan) • Zymark Twister I • H&P Teleshake Electromagnetic Shaker • STarStation enabled Luminex

  10. Engineering Issues • Luminex requires platform >4 inches • Custom plates from Millipore for increased durability and uniformity • Access to the XYP plate holder

  11. Workflow Management • OVERLORD™ • Supervisory software • Controls multiple instruments • Over 140 instruments supported • Use native instrument software for full functionality

  12. Phase 1 System Workflow Samples Worklists Prepare Plates STar-Luminex Incubator Shaker Stack Data Analysis & Reporting

  13. Integration of the Luminex Instrument • STarStation software • Integration Module • Designed for robotics integration • Reliable & Robust

  14. QA – 2 Use Cases Two Scenarios in HTB • Throughput & Accuracy • Standards & Controls on All Plates • Samples run in duplicate or triplicate • Screening for Hits • Standards & Controls on First and Last Plates • Samples run in triplicate & analyzed individually

  15. STarStation Reporting • STarStation reports on a per plate basis • All reports are in xml format • Reports are generated using either the curve on the plate or using a Reference Curve

  16. STarStation Reporting • STarStation tools for analysis • Curve-fit selection • Full control of axes for curve presentation • Levels of Quantification • Automatic outlier rejection

  17. STarStation Reporting

  18. Reporting using Reference Curves • Create a Standard Curve Protocol by selecting the Worklists with the Reference Curves

  19. Reporting using Reference Curves • Adjust Curve-Fits and remove outliers as required

  20. Reporting using Reference Curves • Select Standard Curve Protocol to be used to create results

  21. Reporting using Reference Curves • Select the STarStation Worklists that you want to apply the curve protocol to

  22. Reporting using Reference Curves • Reports on a per plate basis • All relevant Standard Curve information is added to the individual reports

  23. Results!

  24. Future Directions • Applications – Reagents • SNP-based assays • Transcription Factors • Integration • OVERLORD™ Scheduler • Applications – Software • STarStation developments • STarBase

  25. Phase 2 System Workflow Scheduler Samples Worklists Prepare Plates STar-Luminex Incubator Shaker Stack Data Analysis & Reporting

  26. Data Handling • OVERLORD™ Scheduler (Phase 2) • pre-emptive scheduler • can handle multiple critical time points

  27. STarStation Developments • Further automation of instrument procedures • Automated import of Sample information • Expanded repertoire of applications • Example: STarBase allele calling software

  28. Summary • Proof of Principle demonstrated with Phase 1 • STarStation provides reliable & robust integration for the Luminex platform • Phase 2 takes throughput up to 20 plates • Higher throughput and additional applications require more sophisticated analysis tools

  29. Special Thanks • GSK High Throughput Biology • Steve Jeal • Nicola Hendricks • Simon • Wayne Southwick • Martin Whose hard work and patience were the driving force behind this project

  30. More Information • Applied Cytometry Systems • Erin Longdo, VP Business Development • Booth 856 • Process Analysis & Automation • Malcolm Crook, Director Laboratory Automation • Booth 344
