  2. 什么是AMV和M-MLV? • AMV来自禽成纤维病毒逆转录酶,引物延伸反应通常用AMV,AMV性能更好,对次级结构耐受。用AMV的延伸温度可达55℃, 以消除次级结构,但在这样温度下,引物延伸反应的得率通常降低。 • M-MLV来自鼠白血病逆转录酶,M-MLV的最适温度为37℃,在更高温度下,M-MLV不稳定,但M-MLV可合成更长的产物,这一特点对引物延伸反应不适用,因引物延伸反应所得的产物一般较短,为100-500个核苷酸。

  3. AMV和M-MLV的区别(一) • 如果RNA非常复杂,含有二级结构等,AMV保温温度要高,反转录效果要比M-MLV好AMV和M-MLV在本身的聚合酶活性之外,都具有内源RNaseH活性。 • AMV要比M-MLV RNaseH的活性要高-----RNaseH活性同反转录酶的聚合酶活性相互竞争RNA模板与DNA引物或cDNA延伸链间形成的杂合链,并降解RNA:DNA复合物中的RNA链。被RNaseH活性所降解的RNA模板不能再作为合成cDNA的有效底物,降低了cDNA合成的产量和长度,这点来说AMV不如M-MLV效果好。
