its the short and precise data about the blotting and limitations of Slot Blot Technique.<br>There are four types of Blotting Techniques which are following<br>Southern Blotting to identify DNA<br>Northern Blotting to detect RNA<br>Western Blotting to detect proteins<br>These techniques have the different procedures.<br>in Southern, we need to use Restriction enzyme to cut the DNA.<br>Then we need, Gel Electrophoresis and add NAOH base to separate the double strand of DNA into the single strand.<br>then this would be shifted to the membrane and put a nitrocellulose membrane on it.

Difference between slot and dot blot technique • In dot blotting the nucleic acids are blotted as circular blots • In slot blotting they are blotted into rectangular slots. • The difference between dot blot and slot blot procedures is in the way that the nucleic acid mixture is blotted onto the membrane.

  3. Similarities between two techniques • They provide no information about molecular weight. • Dot blots and slot blots are also beneficial when screening a large number of samples, or if a simple answer will suffice. • Useful variation on the typical western blot. • Both provide a quick and efficient means of examining a range of antibody dilutions.

  4. Types of blotting • Southern Blotting (used for analysis of DNA sequence) • Western Blotting (used for detection and analysis of protein in given sample) • Northern Blotting ( Used to analyse and detection of RNA ) • Eastern Blotting ( To identify carbohydrates, epitopes and lipids)

  5. Southern Blotting • Used in phylogenetic analysis. • To identify the gene rearrangements.

  6. Applications of Western and Northern Blotting • Western blotting is used in clinical purposes. Also used to quantifying a gene product and to detect specific protein in low quantity. • Northern blotting is used in screening.

  7. Application of Eastern Blotting • Detection of protein modifications • Used for binding studies. • Used to purify various phospholipids.

  8. Limitations for Slot blot • Signal Normalization • Higher backgrounds • Multiple targets • Laborious • Time consuming
