  1. From Strasbourg... ... but in Köln Very short uptade on Strasbourg analyses UNIVERSITÉ DE STRASBOURG

  2. 1) Comparative adaptability to physical inactivity in bears and humans Bear Captures Inactive - fasting Bed resting Humans Inactive - fed Fasting Humans Active - fasting Köln ongoing study, Analyses april 2011 Constance fast clinic, collaboration initiated Metabolome (U,P) Under analyses Proteome (M,U) Constance fast clinic, collaboration initiated Köln ongoing study, Analyses april 2011 Start Nov 2011 Ox. stress (M,P,A) Under analyses Köln ongoing study Coagulation (M,P,A) Ole/Berolla Köln ongoing study Insulin sig. (M,P,A) Start April 2011 with Bed rest samples Köln ongoing study M: muscle; P: plasma; A: adipose tissue; U: urine

  3. 2) Ecophysiology of bear hibernation Effect of Bear Captures Body temperature rhythms Ecological determinants of hibernation patterns Alina/Stéphane/Akiko/x (Lot of complementary data needed !) Alina/Jons/Akiko/x ongoing Alina/Stéphane/Akiko/x ongoing (need dates of den entry and ambiant temp) Temp. loggers n=4 retrieved n=x implanted Collaboration with Matt Vijayan (Canada) who already developed specific antibody for bear CBP. His student will be in Strasbourg to help in the analyses in beginning December. I suggest co-first lead for Alina and the student. U. cortisol, CBP

  4. 3) What do we aim to do in the discussed Park experiment Overall objective: To understand the mechanisms used by the bears to prevent muscle disuse atrophy and related adaptations Specific questions that we aim to answer: Odd questions… • Is the glyoxylate cycle functional in bears (lipid->glucose)? • Are bears able to synthesis essential amino acids? (based on old hypotheses from R Nelson – so far restricted to plant) Functional questions… • What are the carbon sources for neo-glucogenesis (alanine glycerol) ? • What are the fractional synthesis and catabolic rates of proteins in different organs (previous studies only measured catabolism)? • Is there any between-organ differences? • What is the role of the liver, kidney and gut in glucose production? • What are the organs which participates to the energy saved during hibernation? • What is the insulin sensitivity during hibernation? Methods: Isotope infusions, biopsies, imaging (echography)

  5. 4) Funding plans • Frenche Space Agency: -> will support 2012 activities – amount yet not decided. • 2011 French ANR ‘bioadap’ program: -> More discussions needed with Pierre and Andreas to define common questions (December). • 2012 French ANR ‘white’ program: -> Project focused on true physiological questions almost written (more easy for me than pure ecology stuff as above…). Need to include budget for the Barn. Discussion with Jon-and-Jon and Ole needed. • 2012 French Association for the Study of Myopathies: -> interested to support the project on physiology. They will review proposal in March 2012.

  6. 5) Needed review on “Physiology of hibernation in bears” : who is in? PROPOSITION: • General introduction to the species Jon / Andreas / ? • Biochemistry: Alina / Jon / ? • Energetic: Andrea / Stéphane / ? • Muscle: Stéphane / ? • Cardiovascular: Ole / ? • Immune: Berolla / ? • Bone Ole / ?

  7. 14-hr test on 10 subjects Few pictures from our current bed rest:
