  2. Determenation of Aspartate aminotransferase (AST) • Principle of the method : • Aspartate + 2-oxoglutrate AST Glutamate + Oxalacetate • Oxalacetate + NADH +H+MDHMalate + NAD+

  3. Distribution: • The AST found in highest conc. in heart muscle, liver and skeletal muscle. • Smaller amounts are found in kideny, pancreas and erythrocytes. • Clinical significance • myocardial infarction, hepatocellular disease, and disorders of skeletal museles.

  4. Samples:serum or plasma • Procedure • Assay conditions: • Wavelength ......................... 340nm. • Cuvette ................................ 1cm. light path • constant tempreture .................. 37℃ • Adjust the instrument to zero with distilled water or air.

  5. Pipette into a cuvette: • Mix, incubate at 37℃ for 1 minute. • Read initial absorbance of the sample, start the the stopwach and read absorbances at 1 minute intervals thereafter for 2 minutes. • Calculate the difference between absorbances and the average absorbance differences per minute ( ΔA/min)

  6. CalculationsΔA/min x 1768= U/L of AST • Reference values: • AST: • Men up to 37 U/L • Women up to 31 U/L
