  1. enrichment of DTC in peripheral blood samples of 101 patients with transitional cell carcinoma of the bladder (bTCC); • 208 blood samples(BS)before & after TUR-B, cystectomy (cys) • density gradient centrifugation (Ficoll) by magnetic cells sorting • (AutoMACS, Miltenyi Biotec) • depletion of leukocytes (CD45 microbeads) with • DTC in the negative fraction • Cytospin centrifugation and ICC (anti-CK-ab MNF116, Dako) • DTC detection & quantification by positive CK staining and patho-morphological assessment, FISH (UroVysion) for selected cases • detection of typical chromosomal alterations Detection of disseminated tumor cells (DTC) in the peripheral blood of bladder cancer patients for therapy assessment U. 2. Results/Summary: • 58 of 208 blood samples (28 %) from 33 patients contained DTC • detection of DTC independent of tumor stage & grade • cells of bTCC disseminate early • less DTC after TUR-B & cystectomy compared to pre-operative BS • surgical intervention caused a clear reduction of DTC Detection of disseminated tumor cells (DTC) in the peripheral blood of bladder cancer patients for therapy assessment
