  2. 目的 • 学习离体在位蟾蜍心脏的恒压灌流法 • 观察前,后负荷和心肌收缩性能对心功能的影响,并描绘心功能曲线. • 制备实验性全心衰模型 • 观察强心药物对衰竭心肌的治疗作用.

  3. 原理 • C.O.=S.V. ×HR • HR:离体(心率恒定) • S.V.: • Preload • Afterload • Contractility

  4. 原理 • 心功能不全cardiacinsufficiency 舒缩功能降低或者充盈受限 C.O • Cs2+- Ca2+ 竞争抑制Ca2+内流 • Cardiacglycosides(digitoxin): • Inhibiting Na+/K+ ATPase→ Na+ - Ca2+ ↑

  5. 实验流程 • 1破坏中枢→暴露心脏→分离左右主动脉→分离下腔静脉→结扎,插管 • 2观察 *改变前负荷(储液瓶零点高度),记录C.O.和心率. *改变后负荷(侧管高度),记录C.O.和心率. *改变心肌收缩力: NA, Ach, isoprenaline, propranolol, CsSO4, cardiac glycosides

  6. 注意事项: • 勿用手捏拿心脏,以免损伤心脏。 • 心脏表面滴加任氏液,保持湿润。
