Chromatography : • Definition : Any process which run in coloumn using a fixed between chramatography can be defined as method of seperation of mixture of component into individual components through Equilibrium distribution between two phases. • Classification - • Chromatography is one of the most widely used physico – chemical method seperation of In-organic and organic substance related in composition & properties. • Seperation Mechanism • Technique of carrying out the operation • Medium in which separation is performed.

Types of Paper Chromatography : • Paper partition chromatography • Paper adsorption chromatography • Classification - • Ascending paper chromatography • Descending paper chromatography • Application - • Paper chromatography has been applied to the separation of many organic and biochemical products. • It has been used in the study of inorganic metal salts and complex ions.

  4. Thin layer chromatography : • Thin layer chromatography is similar to paper chromatography. Except that thin (0.25 mm) layer of same inert material. • A layer of any one of these oxides is made from a slurry of powder in suitable inert solvent. • Thin layer chromatography may be used with reagent such as H2SO4 it reacts with paper and is especially useful for the separation and analysis of high molecular weight biochemical compounds. • TLC is often named by other names such as drop, strip, spread layer, surface chromatography and open column chromatography.

  5. Application of thin layer chromatography (TLC): TLC is used as an analytical tool specially inorganic chem. Of separation and it’s applicability in large number of chemical compounds. TLC is also used in identification of compounds in drug biochemical preparation and natural product. TLC is used in checking of distillation fractions and progress of purification by molecular distillation. The high sensitivity of TLC is used to check purity of sample because higher sensitivity enables imparities to be observed in pure sample.

  6. Rf value and Variation – In paper chromatography the result are represented by RF value. Migration of solute relative to the solyent Front. This indicating position of migrated spots on chromatogram. The RF value of calculated Distance travelled by solute Distance travalled by solvent front R indicates fraction of the solute molecule in solvent.
