  1. Department: Physics Intern: AssyaAbourabia Mentor: Dr. Jonathan Celli • Dr. Celli and his team pursue interdisciplinary research on pancreatic cancer. • Dr. Celli research group is focused on problems at the intersection of biophysics and cancer biology. • The basic idea of his approach is to bioengineer the tumor microenvironment so he can put cancer cells back into physiological context. In this way he brings back crucial interactions, and making cancer cells harder to kill in the lab and then treat them with photodynamic therapy (PDT). So that the answers he gets will have more meaning in the clinic.

  2. What is the Autoclave? • Autoclave (steam sterilizer) is a large steel vessel used tosterilize equipment, media, and infectious waste by subjecting them to high pressure saturated steam at 250 °F for around 15 minutes depending on the size of the load and the contents. Gravity Displacement Autoclave • In the gravity displacement autoclave the steam is pumped into the chamber from the upper rear to displace the cold air through the bottom of the chamber. Vacuum Autoclave • In the vacuum autoclave the cold air is removed from the autoclave by a vacuumpump, then steam enters to the steam chamber.

How Does The Killing Occur? • Steam penetrates objects with cooler temperature, which gets condensed to water. • Condensation leads to a decrease in steam volume, which creates negative pressure and draws more steam to the area. • The superheated water molecules rapidly conduct heat into microorganisms, which become so energetic. • The vibratory motion of every molecule of a microorganism is increased to levels that induce the cleavage of intermolecular hydrogen bonds between proteins. • Death is therefore caused by an accumulation of irreversible damage to all metabolic functions of the organism.

  4. Safety Considerations • NEVER try to open while it’s operating. • Wear heat resistant gloves, safety goggles. • Check pressure gauge = 0 psi before opening. • If there’s resistance, do not force. • Stand back. • Open door 1/2 inch, allow steam to dissipate. • Slowly open door further. • Report malfunctions or accidents to your supervisor immediately.
