PRODUKSI FITASE Gen K. pneumoniaASR1 Plasmid (pET22b(+)-Plasmid-Vektor (Novagen) dengan sekuaen His-Tag pada C-Terminal (Novagen) Fermentasi pada fermentor (Stationary Batch Culture): 37oC, 48 jam, media Luria Bertany (Sajidan, 2002) Dikloning pada E. coli BL21 dengan induktor 1 mM IPGT atau Isopropyl-b-D-Thiogalactopyranoside (Novagen) Purifikasi menggunaan Ni-NTA-Affinitas kromatografi (Quiagen) Uji aktivitas dengan melihat warna kuning dari fosfomolibdat dengan spektrofotometer pada l415 nm (Sajidan, 2002) Fitase hasil E. coli rekombinan

Keuntungan Pemakaian Fitase • Penurunan pemakaian Ca dan P anorganik dalam pakan (Abondano, 2005) • Pemanfaatan nutrien lebih baik: protein, asam amino, karbohidrat, lemak, P dan mineral lain (Pallauf dan Rimbach, 1997) • Penurunan biaya pakan (Zanini dan Sazzad, 1999) • P anorganik sekreta rendah, sehingga mencegah pendangkalan sungai (Shin et al., 2001)
