This supplemental figure illustrates the ability of the 35S-SDIR1 transgene to complement the ABA and NaCl insensitive phenotypes of the sdir1-1 mutant (SALK_052702). Panel A shows RNA-blot analysis confirming adequate expression in wild-type, sdir1-1, and transgenic plants, with rRNA as loading control. Panel B presents germination assays on MS media supplemented with NaCl at concentrations of 100 mM and 150 mM, indicating restored viability in transgenic plants. Panel C depicts germination on MS media with 1 mM ABA, demonstrating similar complementation results.

A. RNA-blot analysis of wild-type, sdir1-1 mutant(SALK_052702), WT/35S-SDIR1 and sdir1-1/35S-SDIR1 plants. The rRNA was used as an RNA-loading control. B. Complementation of NaCl insensitive phenotypes of sdir1-1. Wild-type, sdir1-1, WT/35S-SDIR1 and sdir1-1/35S-SDIR1 plants were germinated on MS as controls, and on MS plates with 100 and 150 mM NaCl, respectively, for 6 days. Representative seedlings were selected for photograph. C. Complementation of ABA insensitive phenotypes of sdir1-1. Wild-type, sdir1-1, WT/35S-SDIR1 and sdir1-1/35S-SDIR1 plants were germinated on MS plates and 1 mM ABA for 10 days.
