  4. Embryonic stem cells (ES) are pluripotent cells, that can not only be differentiated to all types of specific adult cells but also can be used for cell therapy. In this survey pluripotent and undifferentiated mouse embryonic stem cells were cultured and surveyed for differentiation into neural and glial cells upon treatment with Nerve Growth Factor

  5. (NGF) .Mouse ES cell line were cultured on inactivated mouse embryonic fibroblasts and the resulting Embryoid Bodies (EBs) were transplanted on the fibronectin and poly-lysine culture plates. The NGF was added to specific neural precursor cells. The expression levels of specific neural genes, including Nestin (marker of neuroepithlial cells), Nkx2.2 ( Ventral neurons in spinal cord marker), nurr1 ( primary dopaminergic neuron(DA) marker), 5100 ( Astrocysts marker) , Olig-2(oligodentrocytes and primary motor neuron marker)by RT -PCR and densitometry were selected to study the differentiation

  7. In addition the formation of dendrites in neurons was assessed through immunocytochemistry assay on microtubule association protein (MAP-2). The express of Nestin and nurr1 proved that NGF cause neural induction and (DA) neurons differentiation. Moreover, we used bFGF together whit NGF synchronically in EBs culture media that showed synergic and special, effects on neural induction and DA neuron differentiation. Some of ES cells under treatment with NGF expressed 5100 and were accordingly proved to be astrocytes. However the expression of Olig2 and Nkx2.2 were not demonstrated and consequently no differentiation into ventral spinal cord neurons and oligodendrocytes, had happened.

  8. Immunocytochemistry assay of MAP-2 confirmed the formation of dendrites and neurons were formed. We conclude that NGF possesses neural induction potential of, ES cells and may be further studied for future therapeutic purposes.
