  1. Synthesis and Characterization of Dendron-based Nanomicelles for Targeted Cancer Therapy Jeromy T. Bentley University of Illinois at Chicago RET Fellow Department of Biopharmaceutical Sciences Advisor: Dr. Seungpyo Hong Mentor: Ryan Pearson June 30, 2011

  2. Impact and Motivation “Different chemo drugs are used to treat different cancers. The drugs go through the whole body to kill the cancer cells.” – American Cancer Society Why do we have to “kill the person to kill the cancer?” How can we alter chemotherapy to provide better treatment of the patient?

  3. Nanomicelles for Passive Targeting Peer et al., Nanocarriers as an emerging platform for cancer therapy. Nature Nanotech. 2007, 2, 751-60.

  4. Summer Plan • Synthesize functionalized hydrophobic polymer “coil” • Conjugate hydrophilic PEGylated dendron to hydrophobic polymer coil to create amphiphilic dendron coil and induced self-assembled structure formation • Characterize synthesized amphiphilic dendron coils and by 1H NMR and FTIR, and nanomicelles by TEM and DLS.

  5. Amphiphilic Dendron coil Hydrophobic coil Hydrophilic mPEGylated-dendron

  6. Nanomicelle

  7. Timeline • Week One: Become familiar with laboratory techniques and research literature • Week Two: Prepare and purify the PLA1oK (up through conjugation with BEI) • Week Three: Prepare and purify the PLA1oK-N3 • Week Four: Prepare and purify the PLA10K-G3 Dendron • Week Five: Prepare and purify the PLA10K-G3 Dendron-mPEG • Week Six: Characterization of Self-Assembled Nanomicelle • Week Six: Preparation of Poster Presentation and Technical Paper • Week Seven: Revisions to Presentation and Final Technical Paper
