Functional equivalence between infection of N. benthamiana by wild type Ourmia melon virus (OuMV) and by agroinfiltration of a mix of three A. tumefaciens strains, each transformed with one of the binary plasmids expressing each of the genomic RNA segments. In all panels, Mock=agroinfiltration with pBin61, Agro= infection by agroinfiltration, Virus= infection by mechanical inoculation of sap from N. benthamiana infected with OuMV VE9 isolate. A) systemic symptoms on infected N. benthamiana, 6 days post infection (dpi). B) Northern analysis using RNA1-specific probes (upper panel), or a mix of the RNA2 and RNA3 probes (central panel). Bottom panel is a total RNA stain by methylen blue C) Western blot analysis from samples collected 7 days after the infection with WT OuMV or agroinfection with a mix of the three genomic clones. Top panel shows the reaction with anti-MP antibodies; central panel shows the reaction with anti-CP antibodies, and the bottom panel shows Coomassie stain for total protein on SDS-PAGE. Arrows point to the position of the MP and CP bands in the Western analysis.
